Summary:
The sources of nucleotide varability of CHD1 genes were investiaged in this study with the impact of the variation on the efficacy of PCR-based sex determination techniques being discussed. In addition, the reliablity of three sets of CHD1 primers for sex determination in 12 native and eight alien species of Hawaii was evaluated while using a comparative approach for sex determination. While using this approach, special attention was given to rare bird when control specimens were not available. These primers combined, provided a reliable means of sex determination in most species (including one of the last po’o-uli), and have produced a valuable reference database for future expanded population-level studies.
