The Hilo Core Genomics Facility provides DNA sequencing
and DNA Fragment Separation and Detection services, available to UH faculty,
visiting researchers, staff, students, as well as other federal, state and private institutions
in the area. Additional services are available for a fee. See our price list for available services and pricing.
The Core Facility offers DNA sequencing services, which can be used to determine nucleotide sequences of DNA samples such as PCR products and plasmids. Samples are prepared by the users (see Sample Preparation Sheet (Word)) and are submitted to the Core Facility, along with a Sample Sheet Submission (Excel). The Facility in turn processes the submitted samples by performing a sequencing reaction with the Big Dye Terminator v3.1 reagents. This is followed by post-sequencing reaction clean-up using BigDye XTerminator Purification Kit, and subsequent sample-loading into the Applied Biosystems (AB) 3500 Genetic Analyzer instrument. The raw data is collected and analyzed by the AB internal software, and the resulting sequence data is then made available to the user.
To sequence small genomes or a number of genes in multiple individuals, the Core Genomics Facility will be offering sequencing services using an Ion Torrent Personal Genome Machine (PGM). This technology provides high through-put sequencing using a chip. For more information on pricing and specific services, contact the Lab Manager.
DNA Fragment Separation and Detection Services
The Core Facility offers DNA Fragment and Detection services for customer-prepared samples. Samples are prepared by the users and are submitted to the Core Facility, along with a Sample Sheet (Word). DNA fragments are separated using the Applied Biosystems 3500 Genetic Analyzer or the Beckman Coulter Ceq 8000 Genetic Analysis System's capillary electrophoresis and are then made available to the user.

Orchid Virus Detection
Services to detect the common orchid RNA viruses Cymbidium mosaic potexvirus (CymMV) and Odontoglossum ringspot tobamovirus (ORSV) are available at the Core Facility. This is especially useful for farmers who need to know if their orchids are contaminated so that they can safely reproduce and sell them. Once an orchid tissue sample from a leaf or a flower is provided, the lab can provide results of the test rapidly. For more information, contact the Lab Manager.
Services to determine the sex of a variety of bird species are offered by the Core Facility. Once a blood sample is provided by the client, results of the test will be available quickly.
